DSIP
One of the longest-studied neuropeptides, originally isolated as a sleep-inducing messenger from the brain of sleeping rabbits, with an inconsistent body of research to this day.
DSIP (delta sleep-inducing peptide) is a nonapeptide first isolated in 1977 by Schoenenberger and Monnier from the venous brain blood of sleeping rabbits and named for its ability to trigger delta-wave EEG activity in that animal model. Over the following decades it was studied in numerous human trials on sleep, the stress axis, and hormone regulation. The originally described sleep-promoting effect has not been reliably reproduced in independent, more rigorous studies.

Evidence
- The original isolation from the brain blood of sleeping rabbits identified the nonapeptide sequence and showed increased delta-wave EEG activity in the animal model following intraventricular administration. Schoenenberger GA, Monnier M 1977, Proceedings of the National Academy of Sciences, Bd. 74(3), S. 1282-1286, PMID 265572
- In a double-blind crossover study in chronic insomnia patients with polysomnographic monitoring over four nights, sleep improvement under DSIP was minor compared to placebo. Monti JM et al. 1987, International Journal of Clinical Pharmacology Research, Bd. 7(2), S. 105-110, PMID 3583493
- A review summarizing three decades of human and animal research concludes that the originally postulated sleep-inducing effect of DSIP could not be consistently confirmed in independent studies. Kovalzon VM, Strekalova TV 2006, Journal of Neurochemistry, Bd. 97(2), S. 303-309, PMID 16539679
- A 2024 preclinical study examined a DSIP fusion peptide in a mouse model of pharmacologically induced insomnia and described effects on neurotransmitter levels such as serotonin and dopamine. Mu X et al. 2024, Frontiers in Pharmacology, Bd. 15, Artikel 1439536, PMID 39444618
Frequently asked questions
Has the sleep-promoting effect of DSIP been confirmed in studies
The evidence is inconsistent. Early studies reported sleep-promoting effects, but a 2006 review concluded that this effect could not be consistently reproduced in independent studies.
Where does the name DSIP come from
DSIP stands for delta sleep-inducing peptide. The name traces back to its first description in 1977, when it was isolated from the brain blood of sleeping rabbits and triggered delta-wave EEG activity in that animal model.
Is DSIP legally available in Germany
DSIP is not approved as a medication in Germany or the EU. It is offered exclusively as a research chemical for laboratory purposes, not for human use.
